The 59-year-old man was on a KTM 1290 Superduke R20 motorbike when the accident happened just before 17:00 on Friday on the B741 southwest of Hamilton Bridge, Crosshill.
He was pronounced dead at the scene and no other cars were involved in the crash.
The road re-opened earlier on Saturday, having been closed while emergency services were at the scene.
Police Scotland Sgt Chris Hoggans said the force’s thoughts were with the family of the man who died.
He added: “We are asking anyone who may have information to contact us.
“If you were on this road around the time of the crash and saw the motorcyclist or if you have dash-cam footage that could assist with our investigation then please contact us.”
